Isolation & characterization of inhibin from human seminal plasma.

Employing a 3 step purifICation procedure follicle stimulat ing hormone (FSH}-suppressing factor, desJgn.lCd "inbibin, w" purified to homogeneity from human IItmin.1 plasma(HSP). The ethanol/etherutract ofHSP was chrorMtoanpbcd 00 Sep~du G-IOO column. The' biologically adive fraction duted was electrophoresed on Pevilum block lODe electropbo~i. and sut-qucntly isolIIted in pure fonn on a OEAE.(:elluIOK column usinl a linear N.CI gradient. The iIolated fK10r wu found to be a non-steroid.l, acidic protein which miSniled as. sinlle band in simple polyacrylamide gels as well •• in SOSpolyacrylamide gels. The susceptibility 10 trypsin and the heat-labi lity of this factor further confirmed the protein nature of illbibill from human IItmioai plasma. From SOS-polyacrylamide JCI electrophoresis tbe molecular weiJht of human seminal plasma illhibiD wu found to be 19.000.
